Vitamin D3 is used as an essential mediator of intestinal calcium absorption and bone calcium metabolism. It is also used as a therapeutic vitamin. Further, it is used as a nutritional supplement and to fortify foods. In addition to this, it is used as pharmaceutical drug and used in the treatment of respiratory diseases involving chronic inflammation such as asthma.
Cholecalciferol is a naturally occuring form of vitamin D. Reported that upon metabolic activation, Cholecalciferol induces cell differentiation and prevents proliferation of cancer cells. Cholecalciferol is an activator of VDR.
Cholecalciferol acts as a hormone precursor since it requires two stages of metabolism: first to 25-hydroxycholecalciferol; then to 1α, 25-dihydroxycholecalciferol. One unit (U.S.P. or international) is defined as the activity of 0.025 μg of vitamin D3 contained in the USP vitamin D reference standard.
Cholecalciferol is sourced either through the diet or exposing skin to ultraviolet rays. Oral administration of vitamin D3 is readily absorbed and is stored in adipose tissue.
Cholecalciferol (Vitamin D3) is known for the maintenance of normal skeleton architecture and mineral homeostasis. Vitamin D3 is synthesized from 7-dehydrocholesterol [CAS: 434-16-2] in the skin under the influence of UV light, or it is obtained from food, especially fatty fish. The transformation (hydroxylation) of vitamin D3 is catalyzed by enzymes in the liver and the kidney to produce calcitriol [C3078] (other name: 1α,25-dihydroxyvitamin D3), the active form of vitamin D3. Calcitriol is active in the regulation of the absorption of calcium from the gastrointestinal tract.
Vitamin D3 is a biologically inactive precursor to calcitriol (Item No. 71820) that is converted to active metabolites in vivo. Vitamin D3 is obtained from dietary sources, including fish, or formed in the epidermis via photolytic conversion of 7-dehydro cholesterol (Item No. 14612) to previtamin D3 by UVB radiation, followed by isomerization to vitamin D3. Vitamin D3 can then be converted to 25-hydroxy vitamin D3 (Item No. 9000683) in the liver by the cytochrome P450 (CYP) isoform CYP2R1 before being converted to calcitriol by CYP27B1 in the kidney. Formulations containing vitamin D3 have been used in the treatment of osteoporosis.
