Forskolin acts as an adenylyl cyclase activator. It is used in the treatment of hypertension, asthma, eczema, psoriasis, congestive heart failure, angina, stomachache, gastric and intestinal spasm. It also plays an important role in the treatment of rheumatism, heart, blood and circulation conditions and cancers. It is used to induce menstruation and as an oral contraceptive.
Forskolin is a diterpenoid isolated from Coleus forskohlii, interacts directly with the catalytic subunit of A cyclase (adenylate cyclase), activating the enzyme and raising intracellular levels of cAMP. Forskolin is cell permeable and active in vivo producing positive inotropic, platelet anti-aggregatory and anti-hypertensive effects in vitro. Has also been shown to exhibit smooth muscle relaxant activity, decrease in intraocular pressure and the promotion of hormonal release from the pituitary glands in vitro. Inhibits MAP kinase, induces phosphorylation of CREB, connexin 43, Bad, ATF-1 and PKB. Forskolin is an inhibitor of AChR.
Forskolin (also called Coleonol) is a diterpene that is produced by the Indian Coleus plant (Coleus forskohlii). Forskolin directly activates adenylyl cyclase (AC) and raises 3',5'-cyclic AMP (cAMP) levels in a wide variety of cell types. AC catalyzes the conversion of ATP to cAMP and pyrophosphate. cAMP is an important second messenger important in many biological processes.
Forskolin, or coleonol, is a cell-permeable, potent, reversible and rapid activator of adenylyl cyclase, an enzyme that converts ATP to cAMP and pyrophosphate (EC50 = 0.5 μM). In combination with 5 other compounds, forskolin enables chemical reprogramming of mouse embryonic fibroblasts to pluripotent stem cells, without genetic factors: Valproic acid, sodium salt (Cat. No. 2815), RepSox (Cat. No. 3742), Tranylcypromine hydrochloride (Cat. No. 3852), CHIR 99021 (Cat. No. 4423) and 3-Deazaneplanocin A hydrochloride (Cat. No. 4703). In combination with IBMX (Cat. No. 2845), forskolin induces neural differentiation of mesenchymal stem cells by downregulating neuron restrictive silencer factor. In a rat model of Huntington's disease-like neurodegeneration, forskolin also exhibits neuroprotective effects. Hypotensive and vasodilatory agent.
